3.4.17.
Sephadex beads, dyed yellow by an immobilzed pH indicator, are used to visualize simulated capture of an analyte (turning red) and  subsequent elution into an external detector (such as AA or MS instrument). Pristine beads (yellow) with an immobilized reagent are loaded into column #1, the sample is aspirated from the flow through port and then perfused through beads in column #1 while matrix passes into waste. Next, the beads are transferred into column #2, analyte is eluted into the
instrument (beads turning yellow), for detection. Finally, the beads are discarded to waste. The movie clip is shown at the speed at which it has been recorded. Red plugs are used to retain the beads. For real life application of this technique see (3.4.19)
